A Reckoning in Blood is part of the main story campaign, Jin's Journey, during Act 2: Retake Castle Shimura.
Synopsis[]
With the shogun contacted, the people of Yarikawa recruited, and the clan Sakai armor back in Jin's possession, Jin Sakai and his allies meet up with Lord Shimura at Shimura's camp to discuss a plan of attack towards their goal of reclaiming Castle Shimura. Shimura has obtained word that Mongol forces lead by Ryuzo have been stationed at Fort Koyasan, and he observes that taking them out would give Shimura an advantage in taking back the Castle. Jin realizes that this is also his chance to confront Ryuzo one last time and end it.
Jin confronts Yuna about joining him to Fort Koyasan, but she refuses, as she is done with the war and simply wants to go to the mainland, and tells Jin that this is his fight, not hers. Jin reluctantly agrees. Taka, however, wants to help, but Jin tells him to listen to his sister. Jin leaves and arrives at the Fort at 7:15 PM, where he goes to scout the location. To Jin's surprise, Taka meets up with him at the fort, where Taka says that he wants to help the Ghost and will serve as a distraction to the Mongols at the entrance. Jin is hesitant, but agrees with the strategy.
Once Taka distracts the guards, Jin makes his way through the fort, where he encounters Ryuzo. Jin prepares to duel Ryuzo, but is ambushed by a Straw Hat and is knocked out. He wakes up, tied to a pole, across from Taka, also captured. Jin asks Taka how he was captured, and Taka replies that once he realized that Jin had been gone for so long, he had to return to the camp to see what happened, resulting in his capture.
Khotun Khan appears and tells Jin that the war can simply be over if Jin tells his forces to surrender. Not only would the war be over, but Jin would be second-in-command to Khotun as he continues with his invasion. Jin refuses the offer. Khotun frees Taka and gives him a sword, ordering him to kill Jin. Taka instead tries to strike at Khotun, but is quickly struck down and beheaded by the Khan. Jin then blacks out from the horror.
Jin wakes up again, at 3:00 PM. Enraged at the death of Taka, Jin frees himself and obtains his armor and weaponry and fights his way to the inner courtyard, where he meets a worried Yuna. Yuna asks where her brother is, and Jin hesitates, causing Yuna to run into the courtyard and discover her brother's decapitated body. Initially, she blames Jin for Taka's death as Taka wanted to be like the Ghost, but the two hear Mongols and Straw Hats closing in on their location and prepare for battle. They kill the arriving forces and escape the courtyard.
At approximately 5:10 PM, Jin and Yuna are honoring Taka's legacy with a grave built for him with his headband atop it. The Tale ends with Jin giving Yuna some time to mourn the loss of her brother.

Preparations are complete for our battle to reclaim Castle Shimura and defeat Khotun Khan. I should speak with my uncle at his staging camp and see if he's ready to launch the attack.
― Jin, before starting the Tale
Ryuzo and his band of Straw Hat traitors are poised to attack our flank if we march to reclaim Castle Shimura. Luckily, I now know his forces are amassing at Fort Koyosan.
I'm going to visit my old friend and end this.
